Conditioned medium from primary cytotrophoblasts, primary placenta-derived mesenchymal stem cells, or sub-cultured placental tissue promoted HUVEC angiogenesis in vitro

Abstract Background As a large capillary network, the human placenta plays an important role throughout pregnancy. Placental vascular development is complex and delicate involves many types of placental cells, such as trophoblasts, mesenchymal stem cells. There has been no systematic, comparative study on roles these two groups cells whole tissue in angiogenesis. In this study, primary cytotrophoblasts (CTBs) from early pregnancy placenta-derived (hPDMSCs) different stages were selected cell research objects, full-term was object to detect effects their conditioned medium (CM) umbilical vein endothelial (HUVEC) Methods We successfully isolated hPDMSCs CTBs, collected CM sub-cultured tissue, then evaluated series angiogenic processes HUVECs vitro. Furthermore, we measured levels factors or by angiogenesis antibody array. Results The results showed that not only but also some extent, promoted HUVEC vitro promoting proliferation, adhesion, migration, invasion, tube formation. found pregnancy, whether CTBs hPDMSCs, played more significant than those cell-derived at 24 h 48 had best effect, tissue-derived 7 days effect among all time points. semiquantitative array 18 43 obvious spots CM, 5 (including CXCL-5, GRO, IL-6, IL-8, MCP-1) highest CM. Conclusions obtained (primary hPDMSCs) contained proangiogenic Therefore, our helpful better understand regulation provides theoretical support for clinical application components, especially engineering treatments.
